Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control HepG2,HeLa,MCF-7,Mouse spleen
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 72kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control HepG2,HeLa,MCF-7,Mouse spleen
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IP Conditions for IP
Recommended dilution 1:500 - 1:1000
Suggested sample / positive control HepG2,HeLa,MCF-7,Mouse spleen
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 72kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene may play a role in regulation of the innate immune response. The encoded protein is upregulated in response to viral infection and may be involved in mediation of tumor necrosis factor-alpha proinflammatory responses. Mutations in this gene have been associated with Aicardi-Goutieres syndrome.
Gene ID 25939
Gene name SAMHD1
Swiss Q9Y3Z3
Synonym SAMHD1,CHBL2,DCIP,HDDC1,MOP-5,SBBI88,SAMHD1 Rabbit pAb,Dendritic cell-derived IFNG-induced protein,Monocyte protein 5,SAM domain and HD domain-containing protein 1,MOP5
Expression Expressed in heart, skeletal muscle, spleen, liver, small intestine, placenta, lung and peripheral blood leukocytes (PubMed:11064105). No expression is seen in brain and thymus (PubMed:11064105).
Functions Host restriction nuclease involved in defense response to virus. Has dNTPase activity and reduces cellular dNTP levels to levels too low for retroviral reverse transcription to occur. Blocks early-stage virus replication in dendritic and other myeloid cells. Likewise, suppresses LINE-1 retrotransposon activity. May play a role in mediating proinflammatory responses to TNF-alpha signaling (PubMed:18546154, PubMed:19525956, PubMed:22056990, PubMed:24336198, PubMed:28229507). Has ribonuclease activity, acting on single-stranded RNA. This activity is essential for H1V-1 restriction (PubMed:25038827).
