Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control Mouse brain
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 109-125kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ICC/IF Conditions for ICC/IF
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control Mouse brain
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Late endosome. Lysosome. Early endosome.
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ELISA Conditions for ELISA
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control Mouse brain
Key conditions Start at the recommended dilution and titrate for your sample and assay

Target data

Background Rubicon plays a role as part of the Beclin1-PI3K complex inhibiting autophagic activity. This protein modulates the fusion of autophagosomes with lysosomes a critical step in degrading cellular components. By preventing excessive autophagy Rubicon helps maintain cellular equilibrium balancing degradation and synthesis processes. Its involvement in endo-lysosomal trafficking is essential for the management of cellular waste and nutrient recycling.
Gene ID 9711
Gene name KIAA0226
Swiss Q92622
Synonym Baron; Beclin 1 associated RUN domain containing protein; Beclin-1 associated RUN domain containing protein; hypothetical protein LOC9711; Kiaa0226; OTTHUMP00000208021; OTTHUMP00000208023; OTTHUMP00000208024; RUBIC_HUMAN
Research area Rubicon also known as Baron is a protein that functions mechanically to regulate autophagy and endosomal maturation. It has a molecular weight of approximately 70 kDa. You can find Rubicon expressed in various tissues but it is especially abundant in the liver spleen and pancreas. The protein acts by interacting with key autophagic and endocytic machinery including Rab7 and UVRAG which modulate vesicular trafficking processes vital for cellular homeostasis.
