Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control Thyroid,Epithelial cell
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 44,KD 27KD ,24 kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control Thyroid,Epithelial cell
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Lysosome, Melanosome, Secreted, extracellular space, Apical cell membrane
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ICC/IF Conditions for ICC/IF
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control Thyroid,Epithelial cell
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Lysosome, Melanosome, Secreted, extracellular space, Apical cell membrane
Controls Include a positive sample, a negative sample, and a secondary or isotype control
FC Conditions for FC
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control Thyroid,Epithelial cell
Key conditions For intracellular targets, optimize fixation and permeabilization, and include an isotype control
Expected result Expect the positive population to shift relative to the negative or isotype control
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background Thiol protease which is believed to participate in intracellular degradation and turnover of proteins (By similarity). Cleaves matrix extracellular phosphoglycoprotein MEPE (By similarity). Involved in the solubilization of cross-linked TG/thyroglobulin in the thyroid follicle lumen (PubMed:12782676). Has also been implicated in tumor invasion and metastasis (By similarity)
Gene ID 3134
Gene name Ctsb
Swiss P10605
Synonym Cathepsin B (YD18146),Cathepsin B (YD18146) Rabbit mAb,Ctsb,Cathepsin B1
Research area CTSB(Cathepsin B) is also named as CPSB and belongs to the peptidase C1 family. It participates in intracellular degradation and turnover of proteins. Cathepsin B precursors found in human malignant ascites fluid do not possess mannose-rich carbohydrates suggesting that a defect in the post translational processing of carbohydrate moieties on tumor. Cathepsin B exists as both glycosylated and unglycosylated forms. In rat macrophages and hepatocytes pro- cathepsin B is 39 kDa (unglycosylated =35 kDa), whereas in human fibroblasts procathepsin B is 44.5-46kDa (unglycosylated=39kDa). It can be detected the 43 kDa form of pro-CTSB, 31 kDa and 25 kDa mature forms in mouse brain by western blot
