Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control Mouse liver
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 45kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control Mouse liver
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Cytoplasm
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background [Isoform 1]: Ca(2+-binding protein that plays a key role in store-operated Ca(2+ entry (SOCE in T-cells by regulating CRAC channel activation. Acts as a cytoplasmic calcium-sensor that facilitates the clustering of ORAI1 and STIM1 at the junctional regions between the plasma membrane and the endoplasmic reticulum upon low Ca(2+ concentration. It thereby regulates CRAC channel activation, including translocation and clustering of ORAI1 and STIM1. Upon increase of cytoplasmic Ca(2+ resulting from opening of CRAC channels, dissociates from ORAI1 and STIM1, thereby destabilizing the ORAI1-STIM1 complex.
Gene ID 84766
Gene name CRACR2A
Swiss Q9BSW2
Synonym CRACR2A,EFCAB4B,CRACR2A Rabbit pAb,Calcium release-activated calcium channel regulator 2A,Calcium release-activated channel regulator 2A,Ras-related protein Rab-46,RAB46
Expression [Isoform 1]: Expressed in the Jurkat T-cell line.; [Isoform 2]: Expressed in endothelial cells (PubMed:25475730). Expressed in Weibel-Palade bodies (which are P-selectin/SELP negative) in endothelial cells (PubMed:31092558). Expressed in the Jurkat T-cell line (PubMed:30814157, PubMed:27016526).
