Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control HepG2,HeLa,MCF7,Rat testis
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 250kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control HepG2,HeLa,MCF7,Rat testis
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IP Conditions for IP
Recommended dilution 1:20 - 1:50
Suggested sample / positive control HepG2,HeLa,MCF7,Rat testis
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 250kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ChIP Conditions for ChIP
Recommended dilution 1:20 - 1:100
Suggested sample / positive control HepG2,HeLa,MCF7,Rat testis
Key conditions Include Input, an IgG negative control, and a positive control for a known binding site
Expected result Expect enrichment at the positive site and low background for the negative site and IgG
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene encodes a TAF (TATA box-binding protein-associated factor), which associates with TBP (TATA box-binding protein) to form the B-TFIID complex that is required for transcription initiation of genes by RNA polymerase II. This TAF has DNA-dependent ATPase activity, which drives the dissociation of TBP from DNA, freeing the TBP to associate with other TATA boxes or TATA-less promoters.
Gene ID 9044
Gene name BTAF1
Swiss O14981
Synonym BTAF1,MOT1,TAF(II)170,TAF172,TAFII170,BTAF1 Rabbit pAb,ATP-dependent helicase BTAF1,B-TFIID transcription factor-associated 170 kDa subunit,TBP-associated factor 172
Functions Regulates transcription in association with TATA binding protein (TBP). Removes TBP from the TATA box in an ATP-dependent manner.
