Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control U-251MG,Rat spleen
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 118kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control U-251MG,Rat spleen
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Cytoplasm,Mitochondrion,Nucleus,Nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control U-251MG,Rat spleen
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Cytoplasm,Mitochondrion,Nucleus,Nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control
FC Conditions for FC
Recommended dilution 1:20 - 1:50
Suggested sample / positive control U-251MG,Rat spleen
Key conditions For intracellular targets, optimize fixation and permeabilization, and include an isotype control
Expected result Expect the positive population to shift relative to the negative or isotype control
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background The protein encoded by this gene catalyzes the first step in ubiquitin conjugation to mark cellular proteins for degradation. This gene complements an X-linked mouse temperature-sensitive defect in DNA synthesis, and thus may function in DNA repair. It is part of a gene cluster on chromosome Xp11.23. Alternatively spliced transcript variants that encode the same protein have been described.
Gene ID 7317
Gene name UBA1
Swiss P22314
Synonym UBA1,UBA1 Rabbit mAb,Protein A1S9,Ubiquitin-activating enzyme E1,A1S9T,UBE1
Expression Detected in erythrocytes (at protein level). Ubiquitous.
Functions Catalyzes the first step in ubiquitin conjugation to mark cellular proteins for degradation through the ubiquitin-proteasome system (PubMed:1606621, PubMed:1447181). Activates ubiquitin by first adenylating its C-terminal glycine residue with ATP, and thereafter linking this residue to the side chain of a cysteine residue in E1, yielding a ubiquitin-E1 thioester and free AMP (PubMed:1447181). Essential for the formation of radiation-induced foci, timely DNA repair and for response to replication stress. Promotes the recruitment of TP53BP1 and BRCA1 at DNA damage sites (PubMed:22456334).
