Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:1000-1:3000
Suggested sample / positive control U-251MG,Mouse liver,Mouse brain
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 250kD
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50-1:200
Suggested sample / positive control U-251MG,Mouse liver,Mouse brain
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation

Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF Conditions for IF
Recommended dilution 1:100-1:800
Suggested sample / positive control U-251MG,Mouse liver,Mouse brain
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Nucleus, Cytoplasm, Chromosome
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IP Conditions for IP
Recommended dilution 1:50-1:200
Suggested sample / positive control U-251MG,Mouse liver,Mouse brain

Target data

Background This gene encodes the largest subunit of RNA polymerase II, the polymerase responsible for synthesizing messenger RNA in eukaryotes. The product of this gene contains a carboxy terminal domain composed of heptapeptide repeats that are essential for polymerase activity. These repeats contain serine and threonine residues that are phosphorylated in actively transcribing RNA polymerase. In addition, this subunit, in combination with several other polymerase subunits, forms the DNA binding domain of the polymerase, a groove in which the DNA template is transcribed into RNA.
Gene ID 5430
Gene name POLR2A
Swiss P24928
Synonym POLR2A,DNA-directed RNA polymerase II subunit RPB1,POLR2,RNA polymerase II subunit B1,DNA-directed RNA polymerase II subunit A,DNA-directed RNA polymerase III largest subunit,RNA-directed RNA polymerase II subunit RPB1,RNA-directed RNA polymerase I
