Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control HL-60,HepG2,SKOV3,MCF7,BxPC-3,Mouse liver,Rat liver
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 68kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control HL-60,HepG2,SKOV3,MCF7,BxPC-3,Mouse liver,Rat liver
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Endoplasmic reticulum membrane,Melanosome,Single-pass type I membrane protein
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene encodes a type I integral membrane protein found only in the rough endoplasmic reticulum. The encoded protein is part of an N-oligosaccharyl transferase complex that links high mannose oligosaccharides to asparagine residues found in the Asn-X-Ser/Thr consensus motif of nascent polypeptide chains. This protein forms part of the regulatory subunit of the 26S proteasome and may mediate binding of ubiquitin-like domains to this proteasome.
Gene ID 6184
Gene name RPN1
Swiss P04843
Synonym RPN1,OST1,RBPH1,RPN1 Rabbit pAb,Dolichyl-diphosphooligosaccharide--protein glycosyltransferase 67 kDa subunit,Ribophorin I,Ribophorin-1
Expression Expressed in all tissues tested.
Functions Essential subunit of the N-oligosaccharyl transferase (OST) complex which catalyzes the transfer of a high mannose oligosaccharide from a lipid-linked oligosaccharide donor to an asparagine residue within an Asn-X-Ser/Thr consensus motif in nascent polypeptide chains.
