Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control 293T,293T+UV,NIH/3T3,C6
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 32kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control 293T,293T+UV,NIH/3T3,C6
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Nucleus,PML body
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IP Conditions for IP
Recommended dilution 1:500 - 1:1000
Suggested sample / positive control 293T,293T+UV,NIH/3T3,C6
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 32kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ChIP Conditions for ChIP
Recommended dilution 1:500 - 1:1000
Suggested sample / positive control 293T,293T+UV,NIH/3T3,C6
Key conditions Include Input, an IgG negative control, and a positive control for a known binding site
Expected result Expect enrichment at the positive site and low background for the negative site and IgG
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background As part of the heterotrimeric replication protein A complex (RPA/RP-A, binds and stabilizes single-stranded DNA intermediates, that form during DNA replication or upon DNA stress. It prevents their reannealing and in parallel, recruits and activates different proteins and complexes involved in DNA metabolism. Thereby, it plays an essential role both in DNA replication and the cellular response to DNA damage. In the cellular response to DNA damage, the RPA complex controls DNA repair and DNA damage checkpoint activation. Through recruitment of ATRIP activates the ATR kinase a master regulator of the DNA damage response. It is required for the recruitment of the DNA double-strand break repair factors RAD51 and RAD52 to chromatin in response to DNA damage. Also recruits to sites of DNA damage proteins like XPA and XPG that are involved in nucleotide excision repair and is required for this mechanism of DNA repair. Plays also a role in base excision repair (BER probably through interaction with UNG. Also recruits SMARCAL1/HARP, which is involved in replication fork restart, to sites of DNA damage. May also play a role in telomere maintenance.
