Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:1000
Suggested sample / positive control THP-1
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 100KDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control THP-1
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Nucleus speckle
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control THP-1
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Nucleus speckle
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IP Conditions for IP
Recommended dilution 1:50 - 1:100
Suggested sample / positive control THP-1
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 100KDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background The removal of introns from nuclear pre-mRNAs occurs on complexes called spliceosomes, which are made up of 4 small nuclear ribonucleoprotein (snRNP) particles and an undefined number of transiently associated splicing factors. This gene product is one of several proteins that associate with U4 and U6 snRNPs. Mutations in this gene are associated with retinitis pigmentosa-18.
Gene ID 9129
Gene name PRPF3
Swiss O43395
Synonym PRPF3,HPRP3,HPRP3P,PRP3,Prp3p,RP18,SNRNP90,PRPF3 Rabbit pAb,Pre-mRNA-splicing factor 3,U4/U6 snRNP 90 kDa protein
Expression Highly expressed in retina, liver, kidney and blood. Detected at lower levels in heart and brain.
Functions Participates in pre-mRNA splicing. Part of the U4/U5/U6 tri-snRNP complex, one of the building blocks of the spliceosome.
