Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:1000
Suggested sample / positive control U-138 MG,HepG2,HeLa
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 35KDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:100 - 1:200
Suggested sample / positive control U-138 MG,HepG2,HeLa
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Cell membrane,Golgi apparatus,Multi-pass membrane protein,trans-Golgi network membrane
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background The protein encoded by this gene is a member of the phosphatidic acid phosphatase (PAP) family. PAPs convert phosphatidic acid to diacylglycerol, and function in de novo synthesis of glycerolipids as well as in receptor-activated signal transduction mediated by phospholipase D. This protein is a membrane glycoprotein localized at the cell plasma membrane. It has been shown to actively hydrolyze extracellular lysophosphatidic acid and short-chain phosphatidic acid. The expression of this gene is found to be enhanced by epidermal growth factor in Hela cells.
Gene ID 8613
Gene name PLPP3
Swiss O14495
Synonym PLPP3,Dri42,LPP3,PAP2B,PPAP2B,VCIP,PPAP2B Rabbit pAb,Lipid phosphate phosphohydrolase 3,PAP2-beta,Phosphatidate phosphohydrolase type 2b,Phosphatidic acid phosphatase 2b,Vascular endothelial growth factor and type I collagen-inducible protein
Expression Ubiquitously expressed (PubMed:9305923, PubMed:12660161). Highly expressed in heart and placenta (PubMed:9305923).
Functions Catalyzes the conversion of phosphatidic acid (PA) to diacylglycerol (DG). In addition it hydrolyzes lysophosphatidic acid (LPA), ceramide-1-phosphate (C-1-P) and sphingosine-1-phosphate (S-1-P). The relative catalytic efficiency is LPA = PA > C-1-P > S-1-P. May be involved in cell adhesion and in cell-cell interactions.
