Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:1000
Suggested sample / positive control Brain
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 29kDa/43kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control Brain
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: cytoplasm, nuclear speck, nucleoplasm, nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background The protein encoded by this gene is a member of the POU-domain transcription factor family and may be involved in maintaining visual system neurons in the retina. The level of the encoded protein is also elevated in a majority of breast cancers, resulting in accelerated tumor growth.
Gene ID 5458
Gene name POU4F2
Swiss Q12837
Synonym POU4F2,BRN3.2,BRN3B,Brn-3b,POU4F2 Rabbit pAb,Brain-specific homeobox/POU domain protein 3B
Expression Expressed in the brain (PubMed:7691107). Expressed in the ganglion cell layer of the retina (PubMed:7691107).
Functions DNA-binding transcriptional regulator and coregulator that recognizes and binds to the consensus octamer binding site 5'-AT[A/T]A[T/A]T[A/T]A-3' in promoter of target genes (PubMed:19266028, PubMed:23805044). Plays a fundamental role in the gene regulatory network essential for retinal ganglion cell (RGC) differentiation. Cooperates with the transcription factor ISL1 to achieve RGC fate specification in the developing retina. Plays also a role in RGC axon formation and guidance by regulating gene expression of specific target genes. Plays a role in TNFSF11-mediated terminal osteoclast differentiation. Binds chromatin at promoter region of target genes (By similarity).
