Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:1000
Suggested sample / positive control HeLa cells, L02 cells, mouse liver tissue, mouse spinal cord, rat liver tissue
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 26kDa/31kDa/35kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control HeLa cells, L02 cells, mouse liver tissue, mouse spinal cord, rat liver tissue
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Cytoplasm
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background The protein encoded by this gene phosphorylates vitamin B6, a step required for the conversion of vitamin B6 to pyridoxal-5-phosphate, an important cofactor in intermediary metabolism. The encoded protein is cytoplasmic and probably acts as a homodimer. Alternatively spliced transcript variants have been described, but their biological validity has not been determined.
Gene ID 8566
Gene name PDXK
Swiss O00764
Synonym PDXK,C21orf124,C21orf97,HEL-S-1a,PKH,PNK,PRED79,pyridoxal kinase,PDXK Rabbit pAb,Pyridoxine kinase
Expression Ubiquitous (PubMed:9099727, PubMed:31187503). Highly expressed in testis (PubMed:9099727).; [Isoform 3]: In adult testis and spermatozoa.
Functions Required for synthesis of pyridoxal-5-phosphate from vitamin B6.
