Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control A-549,HepG2
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 67kDa/82kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF Conditions for IF
Recommended dilution 1:50 - 1:200
Suggested sample / positive control A-549,HepG2
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Chromosome,Cytoplasm,Nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ELISA Conditions for ELISA
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control A-549,HepG2
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 67kDa/82kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene encodes a member of the barr gene family and a regulatory subunit of the condensin complex. This complex is required for the conversion of interphase chromatin into condensed chromosomes. The protein encoded by this gene is associated with mitotic chromosomes, except during the early phase of chromosome condensation. During interphase, the protein has a distinct punctate nucleolar localization. Alternatively spliced transcript variants encoding different proteins have been described.
Gene ID 23397
Gene name NCAPH
Swiss Q15003
Synonym NCAPH,BRRN1,CAP-H,NCAPH Antibody,Barren homolog protein 1,Chromosome-associated protein H,Non-SMC condensin I complex subunit H,XCAP-H homolog,BRRN,CAPH,KIAA0074
Expression Widely expressed at low level. Expressed in proliferating cells.
Functions Regulatory subunit of the condensin complex, a complex required for conversion of interphase chromatin into mitotic-like condense chromosomes. The condensin complex probably introduces positive supercoils into relaxed DNA in the presence of type I topoisomerases and converts nicked DNA into positive knotted forms in the presence of type II topoisomerases.
