Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control 293T,HepG2,MCF7,K-562,NIH/3T3
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 125kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control 293T,HepG2,MCF7,K-562,NIH/3T3
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Nucleus matrix
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control 293T,HepG2,MCF7,K-562,NIH/3T3
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Nucleus matrix
Controls Include a positive sample, a negative sample, and a secondary or isotype control
FC Conditions for FC
Recommended dilution 1:20 - 1:50
Suggested sample / positive control 293T,HepG2,MCF7,K-562,NIH/3T3
Key conditions For intracellular targets, optimize fixation and permeabilization, and include an isotype control
Expected result Expect the positive population to shift relative to the negative or isotype control
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene encodes a nuclear matrix protein, which is proposed to stabilize certain messenger RNA species. Mutations of this gene are associated with distal myopathy 2, which often includes vocal cord and pharyngeal weakness. Alternatively spliced transcript variants, including read-through transcripts composed of the upstream small nucleolar RNA host gene 4 (non-protein coding) and matrin 3 gene sequence, have been identified. Pseudogenes of this gene are located on chromosomes 1 and X.
Gene ID 9782
Gene name MATR3
Swiss P43243
Synonym Matrin 3,Matrin 3 Rabbit mAb,MATR3,KIAA0723
Functions May play a role in transcription or may interact with other nuclear matrix proteins to form the internal fibrogranular network. In association with the SFPQ-NONO heteromer may play a role in nuclear retention of defective RNAs. Plays a role in the regulation of DNA virus-mediated innate immune response by assembling into the HDP-RNP complex, a complex that serves as a platform for IRF3 phosphorylation and subsequent innate immune response activation through the cGAS-STING pathway (PubMed:28712728). May bind to specific miRNA hairpins (PubMed:28431233).
