Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control Mouse liver,Mouse kidney,Mouse heart,Rat liver
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 60kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control Mouse liver,Mouse kidney,Mouse heart,Rat liver
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Golgi apparatus membrane,Single-pass type II membrane protein
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background There are believed to be over 100 different glycosyltransferases involved in the synthesis of protein-bound and lipid-bound oligosaccharides. UDP-N-acetylglucosamine:alpha-3-D-mannoside beta-1,2-N-acetylglucosaminyltransferase I is a medial-Golgi enzyme essential for the synthesis of hybrid and complex N-glycans. The protein, encoded by a single exon, shows typical features of a type II transmembrane protein. The protein is believed to be essential for normal embryogenesis. Several variants encoding the same protein have been found for this gene.
Gene ID 4245
Gene name MGAT1
Swiss P26572
Synonym MGAT1,GLCNAC-TI,GLCT1,GLYT1,GNT-1,GNT-I,GnTI,MGAT,alpha-1,MGAT1 Rabbit pAb,N-glycosyl-oligosaccharide-glycoprotein N-acetylglucosaminyltransferase I,GGNT1
Functions Initiates complex N-linked carbohydrate formation. Essential for the conversion of high-mannose to hybrid and complex N-glycans.
