Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control Mouse brain,Rat brain
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 102kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene encodes a member of a family of membrane-associated E3 ubiquitin ligases containing RING-CH-type zinc finger motifs. Ubiquitination of type II deiodinase by the encoded protein is an important regulatory step in thyroid hormone signalling. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene.
Gene ID 10299
Gene name MARCH6
Swiss O60337
Synonym MARCH6,DOA10,MARCH-VI,RNF176,TEB4,MARCH6 Rabbit pAb,Doa10 homolog,Membrane-associated RING finger protein 6,Membrane-associated RING-CH protein VI,Protein TEB-4,RING finger protein 176,RING-type E3 ubiquitin transferase MARCHF6,KIAA0597,44261
Expression Present in brain (at protein level).
Functions E3 ubiquitin-protein ligase that promotes ubiquitination of DIO2, leading to its degradation. E3 ubiquitin ligases accept ubiquitin from an E2 ubiquitin-conjugating enzyme in the form of a thioester and then directly transfer the ubiquitin to targeted substrates. May cooperate with UBE2G1.
