Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control BT-474,THP-1,Mouse thymus
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 150kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control BT-474,THP-1,Mouse thymus
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IP Conditions for IP
Recommended dilution 1:20 - 1:50
Suggested sample / positive control BT-474,THP-1,Mouse thymus
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 150kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene is a member of the Jumonji domain 2 (JMJD2) family and encodes a protein containing a JmjN domain, a JmjC domain, a JD2H domain, two TUDOR domains, and two PHD-type zinc fingers. This nuclear protein functions as a trimethylation-specific demethylase, converting specific trimethylated histone residues to the dimethylated form, and as a transcriptional repressor.
Gene ID 9682
Gene name KDM4A
Swiss O75164
Synonym KDM4A,KDM4A Rabbit mAb,JmjC domain-containing histone demethylation protein 3A,Jumonji domain-containing protein 2A,[histone H3]-trimethyl-L-lysine(36) demethylase 4A,[histone H3]-trimethyl-L-lysine(9) demethylase 4A,JHDM3A,JMJD2,JMJD2A,KIAA0677
Expression Ubiquitous.
Functions Histone demethylase that specifically demethylates 'Lys-9' and 'Lys-36' residues of histone H3, thereby playing a central role in histone code (PubMed:26741168). Does not demethylate histone H3 'Lys-4', H3 'Lys-27' nor H4 'Lys-20'. Demethylates trimethylated H3 'Lys-9' and H3 'Lys-36' residue, while it has no activity on mono- and dimethylated residues. Demethylation of Lys residue generates formaldehyde and succinate. Participates in transcriptional repression of ASCL2 and E2F-responsive promoters via the recruitment of histone deacetylases and NCOR1, respectively.
