Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control Use a sample with high target expression as the positive control
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 21kD ,54 kDa, 37 kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control Use a sample with high target expression as the positive control
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected staining follows the target localization
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ICC/IF Conditions for ICC/IF
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control Use a sample with high target expression as the positive control
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence follows the target localization
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background IL-12 and IL-23 are heterodimeric cytokines with a common p40 subunit (PMID: 11114383). IL-12 is composed of the IL-12 p40 subunit linked to the IL-12 p35 subunit, and the heterodimer signals through the IL-12 receptor (IL-12R), which comprises the IL-12Rβ1 and IL-12Rβ2 subunits. IL-23 is composed of the IL-23 p19 subunit and the IL-12 p40 (IL-12/23p40) subunit, which signals through IL-23R and IL-12Rβ1 (PMID: 11114383; 26121196 ). The importance of IL-23 has been confirmed by clinical data, as monoclonal antibodies (MABs) targeting the p40 subunit (shared by IL-23 and IL-12), as well as p19 (specific for IL-23) were effective in therapy for psoriasis and psoriatic arthritis (PMID: 25754330; 18486740). This antibody can recognize the IL12B monomer (37 kDa) and the heterodimer (IL12B+IL23A, 54 kDa).
Gene ID 51561
Gene name IL23A
Swiss Q9NPF7
Synonym IL 23, IL23, IL23 (IL12B+IL23A), IL23(IL12B+IL23A)
Research area IL12B to form the IL-23 interleukin , an heterodimeric cytokine which functions in innate and adaptive immunity. IL-23 may constitute with IL-17 an acute response to infection in peripheral tissues. IL-23 binds to an heterodimeric receptor complex composed of IL12RB1 and IL23R , activates the Jak-Stat signaling cascade , stimulates memory rather than naive T-cells and promotes production of proinflammatory cytokines. IL-23 induces autoimmune inflammation and thus may be responsible for autoimmune inflammatory diseases and may be important for tumorigenesis. ,induction:Up-regulated by a wide array of pathogens and pathogen-products together with self-signals for danger or injury. Up-regulated in psoriatic dermal tissues , in dendritic cells of multiple sclerosis patients and in tumors. ,similarity:Belongs to the IL-6 superfamily. ,subcellular location:Secreted upon association with IL12B. ,subunit:Heterodimer with IL12B; disulfide-linked. The heterodimer is known as interleukin IL-23. ,tissue specificity:Secreted by activated dendritic and phagocytic cells and keratinocytes. Also expressed by dermal Langerhans cells (at protein level) . ,
