Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control HeLa,NIH/3T3
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 15kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control HeLa,NIH/3T3
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: cytosol,nucleoplasm,nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IP Conditions for IP
Recommended dilution 1:20 - 1:50
Suggested sample / positive control HeLa,NIH/3T3
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 15kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
FC Conditions for FC
Recommended dilution 1:20 - 1:50
Suggested sample / positive control HeLa,NIH/3T3
Key conditions For intracellular targets, optimize fixation and permeabilization, and include an isotype control
Expected result Expect the positive population to shift relative to the negative or isotype control
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ICC/IF Conditions for ICC/IF
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control HeLa,NIH/3T3

Target data

Background Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a replication-dependent histone that is a member of the histone H2B family. Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element. This gene is found in the large histone gene cluster on chromosome 6p22-p21.3.

Functions Core component of nucleosome. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling.
