Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control HeLa,HepG2,Raji,Mouse thymus,Rat lung
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 29kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control HeLa,HepG2,Raji,Mouse thymus,Rat lung
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Chromosome,Nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control HeLa,HepG2,Raji,Mouse thymus,Rat lung
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Chromosome,Nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ChIP Conditions for ChIP
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control HeLa,HepG2,Raji,Mouse thymus,Rat lung
Key conditions Include Input, an IgG negative control, and a positive control for a known binding site
Expected result Expect enrichment at the positive site and low background for the negative site and IgG
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background Histones are basic nuclear proteins responsible for nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. This gene is intronless and encodes a replication-dependent histone that is a member of the histone H1 family. Transcripts from this gene lack polyA tails but instead contain a palindromic termination element. This gene is found in the large histone gene cluster on chromosome 6.
Gene ID 3006
Gene name HIST1H1C
Swiss P16403
Synonym Histone H1.2,Histone H1.2 Rabbit mAb,HIST1H1C,Histone H1c,Histone H1d,Histone H1s-1,H1F2
Functions Histone H1 protein binds to linker DNA between nucleosomes forming the macromolecular structure known as the chromatin fiber. Histones H1 are necessary for the condensation of nucleosome chains into higher-order structured fibers. Acts also as a regulator of individual gene transcription through chromatin remodeling, nucleosome spacing and DNA methylation (By similarity).
