Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:1000
Suggested sample / positive control HeLa,HepG2
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 33KDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control HeLa,HepG2
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Cytoplasmic side,Endoplasmic reticulum membrane,Microsome,Peripheral membrane protein
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background Heme oxygenase, an essential enzyme in heme catabolism, cleaves heme to form biliverdin, which is subsequently converted to bilirubin by biliverdin reductase, and carbon monoxide, a putative neurotransmitter. Heme oxygenase activity is induced by its substrate heme and by various nonheme substances. Heme oxygenase occurs as 2 isozymes, an inducible heme oxygenase-1 and a constitutive heme oxygenase-2. HMOX1 and HMOX2 belong to the heme oxygenase family.
Gene ID 3162
Gene name HMOX1
Swiss P09601
Synonym HMOX1,HMOX1D,HO-1,HSP32,bK286B10,heme oxygenase 1,Heme Oxygenase 1 (HO-1/HMOX1) Rabbit pAb,HO,HO1
Expression Expressed at higher levels in renal cancer tissue than in normal tissue (at protein level).
Functions Heme oxygenase cleaves the heme ring at the alpha methene bridge to form biliverdin. Biliverdin is subsequently converted to bilirubin by biliverdin reductase. Under physiological conditions, the activity of heme oxygenase is highest in the spleen, where senescent erythrocytes are sequestrated and destroyed. Exhibits cytoprotective effects since excess of free heme sensitizes cells to undergo apoptosis.
