Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:1000-1:3000
Suggested sample / positive control Raji,Mouse intestine
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 37kD
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:300-1:2000
Suggested sample / positive control Raji,Mouse intestine
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Cell membrane, Endoplasmic reticulum membrane, Early endosome membrane, Late endosome membrane, Lysosome membrane, Autolysosome membrane
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF Conditions for IF
Recommended dilution 1:100-1:800
Suggested sample / positive control Raji,Mouse intestine
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Cell membrane, Endoplasmic reticulum membrane, Early endosome membrane, Late endosome membrane, Lysosome membrane, Autolysosome membrane
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ELISA Conditions for ELISA
Recommended dilution 1:5000-1:20000
Suggested sample / positive control Raji,Mouse intestine
Key conditions Start at the recommended dilution and titrate for your sample and assay

Target data

Background HLA-DRA is one of the HLA class II alpha chain paralogues. This class II molecule is a heterodimer consisting of an alpha and a beta chain, both anchored in the membrane. It plays a central role in the immune system by presenting peptides derived from extracellular proteins. Class II molecules are expressed in antigen presenting cells (APC: B lymphocytes, dendritic cells, macrophages). The alpha chain is approximately 33-35 kDa and its gene contains 5 exons. Exon 1 encodes the leader peptide, exons 2 and 3 encode the two extracellular domains, and exon 4 encodes the transmembrane domain and the cytoplasmic tail. DRA does not have polymorphisms in the peptide binding part and acts as the sole alpha chain for DRB1, DRB3, DRB4 and DRB5.
Gene ID 3122
Gene name HLA-DRA HLA-DRA1
Swiss P01903
Synonym HLA-DRA HLA-DRA1;HLA class II histocompatibility antigen, DR alpha chain (MHC class II antigen DRA);
