Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control Rat liver
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 122kD
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control Rat liver
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Cell projection,Cytoplasm,Nucleus,Perikaryon, axon, dendrite
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ELISA Conditions for ELISA
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control Rat liver
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 122kD
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background Histones play a critical role in transcriptional regulation, cell cycle progression, and developmental events. Histone acetylation/deacetylation alters chromosome structure and affects transcription factor access to DNA. The protein encoded by this gene belongs to class II of the histone deacetylase/acuc/apha family. It contains an internal duplication of two catalytic domains which appear to function independently of each other. This protein possesses histone deacetylase activity and represses transcription.
Gene ID 10013
Gene name HDAC6
Swiss Q9UBN7
Synonym HDAC6,CPBHM,HD6,JM21,PPP1R90,HDAC5 (Phospho-Ser259) Antibody,E3 ubiquitin-protein ligase HDAC6,Tubulin-lysine deacetylase HDAC6,KIAA0901,HDAC5 (Phospho-Ser259)
Functions Responsible for the deacetylation of lysine residues on the N-terminal part of the core histones (H2A, H2B, H3 and H4). Histone deacetylation gives a tag for epigenetic repression and plays an important role in transcriptional regulation, cell cycle progression and developmental events. Histone deacetylases act via the formation of large multiprotein complexes (By similarity). Plays a central role in microtubule-dependent cell motility via deacetylation of tubulin. Involved in the MTA1-mediated epigenetic regulation of ESR1 expression in breast cancer.
