Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control U-251MG,SW480
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 109kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control U-251MG,SW480
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Cytoplasm,Secreted, cytoskeleton
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background The protein encoded by this gene binds to the 'plus' ends of actin monomers and filaments to prevent monomer exchange. The encoded calcium-regulated protein functions in both assembly and disassembly of actin filaments. Defects in this gene are a cause of familial amyloidosis Finnish type (FAF). Multiple transcript variants encoding several different isoforms have been found for this gene.

Gene name GSN
Swiss P06396
Synonym GSN,ADF,AGEL,gelsolin,Gelsolin Rabbit pAb,Actin-depolymerizing factor,Brevin
Expression Phagocytic cells, platelets, fibroblasts, nonmuscle cells, smooth and skeletal muscle cells.
Functions Calcium-regulated, actin-modulating protein that binds to the plus (or barbed) ends of actin monomers or filaments, preventing monomer exchange (end-blocking or capping). It can promote the assembly of monomers into filaments (nucleation) as well as sever filaments already formed. Plays a role in ciliogenesis.
