Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control SKOV3,Mouse brain,Mouse skeletal muscle,Mouse kidney,Rat brain
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 21kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IP Conditions for IP
Recommended dilution 1:20 - 1:50
Suggested sample / positive control SKOV3,Mouse brain,Mouse skeletal muscle,Mouse kidney,Rat brain
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 21kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
FC Conditions for FC
Recommended dilution 1:20 - 1:50
Suggested sample / positive control SKOV3,Mouse brain,Mouse skeletal muscle,Mouse kidney,Rat brain
Key conditions For intracellular targets, optimize fixation and permeabilization, and include an isotype control
Expected result Expect the positive population to shift relative to the negative or isotype control
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ICC/IF Conditions for ICC/IF
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control SKOV3,Mouse brain,Mouse skeletal muscle,Mouse kidney,Rat brain
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Cell membrane,Cytoplasm,Cytoplasmic side,Golgi apparatus,Golgi apparatus membrane,Lipid-anchor,Lipid-anchor,Nucleus,perinuclear region
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene belongs to the Ras oncogene family, whose members are related to the transforming genes of mammalian sarcoma retroviruses. The products encoded by these genes function in signal transduction pathways. These proteins can bind GTP and GDP, and they have intrinsic GTPase activity. This protein undergoes a continuous cycle of de- and re-palmitoylation, which regulates its rapid exchange between the plasma membrane and the Golgi apparatus. Mutations in this gene cause Costello syndrome, a disease characterized by increased growth at the prenatal stage, growth deficiency at the postnatal stage, predisposition to tumor formation, mental retardation, skin and musculoskeletal abnormalities, distinctive facial appearance and cardiovascular abnormalities. Defects in this gene are implicated in a variety of cancers, including bladder cancer, follicular thyroid cancer, and oral squamous cell carcinoma. Multiple transcript variants, which encode different isoforms, have been identified for this gene.

Expression Widely expressed.
Functions Involved in the activation of Ras protein signal transduction (PubMed:22821884). Ras proteins bind GDP/GTP and possess intrinsic GTPase activity (PubMed:12740440, PubMed:14500341, PubMed:9020151).
