Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:1000
Suggested sample / positive control Mouse skeletal muscle,Mouse heart,Rat skeletal muscle
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 28KDa/43KDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:100
Suggested sample / positive control Mouse skeletal muscle,Mouse heart,Rat skeletal muscle
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Secreted
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene encodes a secreted ligand of the TGF-beta (transforming growth factor-beta) superfamily of proteins. Ligands of this family bind various TGF-beta receptors leading to recruitment and activation of SMAD family transcription factors that regulate gene expression. The encoded preproprotein is proteolytically processed to generate each subunit of the disulfide-linked homodimer. This protein negatively regulates skeletal muscle cell proliferation and differentiation. Mutations in this gene are associated with increased skeletal muscle mass in humans and other mammals.
Gene ID 2660
Gene name MSTN
Swiss O14793
Synonym MSTN,GDF8,MSLHP,myostatin,GDF-8/Myostatin Rabbit pAb
Functions Acts specifically as a negative regulator of skeletal muscle growth.
