Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control Mouse brain,Mouse skeletal muscle,Mouse pancreas,Rat brain,Rat skeletal muscle
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 70kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control Mouse brain,Mouse skeletal muscle,Mouse pancreas,Rat brain,Rat skeletal muscle
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Cell junction,Cell membrane,Multi-pass membrane protein, postsynaptic cell membrane, synapse
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background GABA is the major inhibitory neurotransmitter in the mammalian brain where it acts at GABA-A receptors, which are ligand-gated chloride channels. Chloride conductance of these channels can be modulated by agents such as benzodiazepines that bind to the GABA-A receptor. At least 16 distinct subunits of GABA-A receptors have been identified. Transcript variants utilizing three different alternative non-coding first exons have been described.
Gene ID 2558
Gene name GABRA5
Swiss P31644
Synonym GABA A Receptor alpha 5,GABA A Receptor alpha 5 Rabbit mAb,GABRA5,GABA(A) receptor subunit alpha-5
Functions GABA, the major inhibitory neurotransmitter in the vertebrate brain, mediates neuronal inhibition by binding to the GABA/benzodiazepine receptor and opening an integral chloride channel.
