Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control rat brain, rat heart
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 23kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control rat brain, rat heart
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: endoplasmic reticulum, extracellular region, plasma membrane
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene encodes a secreted protein that is released from muscle cells during exercise. The encoded protein may participate in the development of brown fat. Translation of the precursor protein initiates at a non-AUG start codon at a position that is conserved as an AUG start codon in other organisms. Alternative splicing results in multiple transcript variants.
Gene ID 252995
Gene name FNDC5
Swiss Q8NAU1
Synonym FRCP2,irisin,FNDC5,FNDC5 Rabbit pAb,Fibronectin type III repeat-containing protein 2
Expression Widely expressed, with highest levels in heart. Very low expression, if any, in colon, pancreas and spleen.
Functions Irisin: Contrary to mouse, may not be involved in the beneficial effects of muscular exercise, nor in the induction of browning of human white adipose tissue.
