Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control MCF7,Mouse liver
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 36kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control MCF7,Mouse liver
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: cytoplasm, cytosol, extracellular exosome, nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IP Conditions for IP
Recommended dilution 1:20 - 1:50
Suggested sample / positive control MCF7,Mouse liver
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 36kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
FC Conditions for FC
Recommended dilution 1:20 - 1:50
Suggested sample / positive control MCF7,Mouse liver
Key conditions For intracellular targets, optimize fixation and permeabilization, and include an isotype control
Expected result Expect the positive population to shift relative to the negative or isotype control
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background Fructose-1,6-bisphosphatase 1, a gluconeogenesis regulatory enzyme, catalyzes the hydrolysis of fructose 1,6-bisphosphate to fructose 6-phosphate and inorganic phosphate. Fructose-1,6-diphosphatase deficiency is associated with hypoglycemia and metabolic acidosis.
Gene ID 2203
Gene name FBP1
Swiss P09467
Synonym FBP1,FBP1 Rabbit mAb,D-fructose-1,6-bisphosphate 1-phosphohydrolase 1,Liver FBPase,FBP
Expression Expressed in pancreatic islets.
Functions Catalyzes the hydrolysis of fructose 1,6-bisphosphate to fructose 6-phosphate in the presence of divalent cations, acting as a rate-limiting enzyme in gluconeogenesis. Plays a role in regulating glucose sensing and insulin secretion of pancreatic beta-cells. Appears to modulate glycerol gluconeogenesis in liver. Important regulator of appetite and adiposity; increased expression of the protein in liver after nutrient excess increases circulating satiety hormones and reduces appetite-stimulating neuropeptides and thus seems to provide a feedback mechanism to limit weight gain.
