Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control HeLa,Mouse lung,Rat lung
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 270kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control HeLa,Mouse lung,Rat lung
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Cell junction,Cornified envelope,Cytoplasm, cytoskeleton, desmosome
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene encodes a member of the plakin family of proteins that forms a component of desmosomes and the epidermal cornified envelope. This gene is located in the tylosis oesophageal cancer locus on chromosome 17q25, and its deletion is associated with both familial and sporadic forms of oesophageal squamous cell carcinoma. Patients suffering from the autoimmune mucocutaneous disorder, paraneoplastic pemphigus, develop antibodies against the encoded protein.
Gene ID 2125
Gene name EVPL
Swiss Q92817
Synonym EVPL,EVPK,EVPL Rabbit pAb,210 kDa cornified envelope precursor protein,210 kDa paraneoplastic pemphigus antigen,p210
Expression Exclusively expressed in stratified squamous epithelia.
Functions Component of the cornified envelope of keratinocytes. May link the cornified envelope to desmosomes and intermediate filaments.
