Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:200 - 1:2000
Suggested sample / positive control Hela,HT-29,Mouse brain,Mouse eye,Mouse small intestine,Rat brain
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 80kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control Hela,HT-29,Mouse brain,Mouse eye,Mouse small intestine,Rat brain
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Cell membrane,Single-pass type I membrane protein
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background DNER (Delta/Notch-like EGF-related receptor), also known as BET (brain-specific EGF-like transmembrane protein), is a type I transmembrane glycoprotein of the Notch/Delta family . In the mouse, DNER has been detected as 90, 120 and 150 kDa forms which are probably variably glycosylated .Activator of the NOTCH1 pathway. May mediate neuron-glia interaction during astrocytogenesis.DNER associates with protein tyrosine phosphatase zeta (PTP zeta ), which is the receptor of pleiotrophin (PTN). PTP zeta -PTN-DNER signaling has been implicated in the regulation of neuritogenesis. Expression of DNER in glioblastoma stem-like cells inhibits formation of neurospheres in vitro, while in vivo it induces differentiation and inhibits growth of xenografts, thus acting as a tumor suppressor.
Gene ID 92737
Gene name DNER
Swiss Q8NFT8
Synonym DNER,UNQ26,bet,DNER Rabbit pAb
Expression Expressed in brain, spinal cord and adrenal gland.
Functions Activator of the NOTCH1 pathway. May mediate neuron-glia interaction during astrocytogenesis (By similarity).
