Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:2000 - 1:9000
Suggested sample / positive control HeLa,293T
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 18KDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This antimicrobial gene encodes a secreted protein that is subsequently processed into mature peptides of distinct biological activities. The C-terminal peptide is constitutively expressed in sweat and has antibacterial and antifungal activities. The N-terminal peptide, also known as diffusible survival evasion peptide, promotes neural cell survival under conditions of severe oxidative stress. A glycosylated form of the N-terminal peptide may be associated with cachexia (muscle wasting) in cancer patients. Alternative splicing results in multiple transcript variants encoding different isoforms.
Gene ID 117159
Gene name DCD
Swiss P81605
Synonym DCD,AIDD,DCD-1,DSEP,HCAP,PIF,dermcidin,DCD Rabbit pAb,Preproteolysin
Expression Specifically and constitutively expressed in eccrine sweat gland cells (at protein level). Secreted into the sweat at a concentration of 1-10 micrograms/ml.
Functions DCD-1 displays antimicrobial activity thereby limiting skin infection by potential pathogens in the first few hours after bacterial colonization. Highly effective against E.coli, E.faecalis, S.aureus and C.albicans. Optimal pH and salt concentration resemble the conditions in sweat. Also exhibits proteolytic activity, cleaving on the C-terminal side of Arg and, to a lesser extent, Lys residues (PubMed:17448443).
