Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control 293T,LO2,HeLa,Mouse heart,Rat testis
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 54kDa/58kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF Conditions for IF
Recommended dilution 1:50 - 1:200
Suggested sample / positive control 293T,LO2,HeLa,Mouse heart,Rat testis
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: cytosol, extracellular exosome, nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ELISA Conditions for ELISA
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control 293T,LO2,HeLa,Mouse heart,Rat testis
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 54kDa/58kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene is a member of the family of dihydroxyacetone kinases, which have a protein structure distinct from other kinases. The product of this gene phosphorylates dihydroxyacetone, and also catalyzes the formation of riboflavin 4',5'-phosphate (aka cyclin FMN) from FAD. Several alternatively spliced transcript variants have been found for this gene.
Gene ID 26007
Gene name TKFC
Swiss Q3LXA3
Synonym TKFC,DAK,NET45,DAK Antibody,Bifunctional ATP-dependent dihydroxyacetone kinase/FAD-AMP lyase (cyclizing),Glycerone kinase,Triokinase,Triose kinase,FAD-AMP lyase (cyclic FMN forming),FMN cyclase
Expression Detected in erythrocytes (at protein level).
Functions Catalyzes both the phosphorylation of dihydroxyacetone and of glyceraldehyde, and the splitting of ribonucleoside diphosphate-X compounds among which FAD is the best substrate. Represses IFIH1-mediated cellular antiviral response (PubMed:17600090).
