Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control Epithelial cell
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 48kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC-P Conditions for IHC-P
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control Epithelial cell
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Nucleus matrix, Cytoplasm, perinuclear region, Nucleus, nucleolus
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background Required for the formation of KRT8/KRT18 filaments that are involved in ARHGEF40-mediated actin stress fiber formation and tensional force-induced stress fiber formation and reinforcement (By similarity). Also acts downstream of ROCK kinase activation as part of a positive feedback mechanism in response to cellular mechanical stress loading (By similarity). Organization and orientation of KRT18 filaments are responsible for the properly elongated morphology of epithelial tubules (By similarity). Involved in the uptake of thrombin-antithrombin complexes by hepatic cells (By similarity). When phosphorylated, plays a role in filament reorganization. Involved in the delivery of mutated CFTR to the plasma membrane. Together with KRT8, is involved in interleukin-6 (IL-6)-mediated barrier protection (PubMed:17213200)
