Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control K-562,BT-474,HeLa,Mouse testis
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 88kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control K-562,BT-474,HeLa,Mouse testis
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: nucleoplasm,nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ICC Conditions for ICC
Recommended dilution 1:50 - 1:200

Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: nucleoplasm, nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IP Conditions for IP
Recommended dilution 1:20 - 1:50
Suggested sample / positive control K-562,BT-474,HeLa,Mouse testis
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 88kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
FC Conditions for FC
Recommended dilution 1:20 - 1:50
Suggested sample / positive control K-562,BT-474,HeLa,Mouse testis
Key conditions For intracellular targets, optimize fixation and permeabilization, and include an isotype control
Expected result Expect the positive population to shift relative to the negative or isotype control
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background CUL4A is the ubiquitin ligase component of a multimeric complex involved in the degradation of DNA damage-response proteins (Liu et al., 2009 [PubMed 19481525]).[supplied by OMIM, Oct 2009]
Gene ID 8451
Gene name CUL4A
Swiss Q13619
Synonym Cullin4A,Cullin4A Rabbit mAb,CUL4A
Functions Core component of multiple cullin-RING-based E3 ubiquitin-protein ligase complexes which mediate the ubiquitination of target proteins. As a scaffold protein may contribute to catalysis through positioning of the substrate and the ubiquitin-conjugating enzyme. The E3 ubiquitin-protein ligase activity of the complex is dependent on the neddylation of the cullin subunit and is inhibited by the association of the deneddylated cullin subunit with TIP120A/CAND1. The functional specificity of the E3 ubiquitin-protein ligase complex depends on the variable substrate recognition component. DCX(DET1-COP1) directs ubiquitination of JUN. DCX(DDB2) directs ubiquitination of XPC. DCX(DDB2) ubiquitinates histones H3-H4 and is required for efficient histone deposition during replication-coupled (H3.1) and replication-independent (H3.3) nucleosome assembly, probably by facilitating the transfer of H3 from ASF1A/ASF1B to other chaperones involved in histone deposition. DCX(DTL) plays a role in PCNA-dependent polyubiquitination of CDT1 and MDM2-dependent ubiquitination of TP53 in response to radiation-induced DNA damage and during DNA replication. In association with DDB1 and SKP2 probably is involved in ubiquitination of CDKN1B/p27kip. Is involved in ubiquitination of HOXA9. DCX(DTL) directs autoubiquitination of DTL.
