Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control A-549,MCF7,Mouse brain,Mouse kidney
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 49kDa/53kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF Conditions for IF
Recommended dilution 1:50 - 1:200
Suggested sample / positive control A-549,MCF7,Mouse brain,Mouse kidney
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Cytoplasmic vesicle,Nucleus,Peripheral membrane protein,Secreted, secretory vesicle membrane
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ELISA Conditions for ELISA
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control A-549,MCF7,Mouse brain,Mouse kidney
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 49kDa/53kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene encodes a member of the M14 family of metallocarboxypeptidases. The encoded preproprotein is proteolytically processed to generate the mature peptidase. This peripheral membrane protein cleaves C-terminal amino acid residues and is involved in the biosynthesis of peptide hormones and neurotransmitters, including insulin. This protein may also function independently of its peptidase activity, as a neurotrophic factor that promotes neuronal survival, and as a sorting receptor that binds to regulated secretory pathway proteins, including prohormones. Mutations in this gene are implicated in type 2 diabetes.
Gene ID 1363
Gene name CPE
Swiss P16870
Synonym CPE,CPH,CPE Antibody,Carboxypeptidase H,Enkephalin convertase,Prohormone-processing carboxypeptidase
Functions Removes residual C-terminal Arg or Lys remaining after initial endoprotease cleavage during prohormone processing. Processes proinsulin.
