Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:100 - 1:500
Suggested sample / positive control 293T,Mouse lung,Mouse testis,Mouse brain,Rat lung
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 37KDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control 293T,Mouse lung,Mouse testis,Mouse brain,Rat lung
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: actin cytoskeleton, cytoskeleton, cytosol, focal adhesion
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene encodes a protein with a markedly acidic C terminus; the basic N-terminus is highly homologous to the N-terminus of a related gene, CNN1. Members of the CNN gene family all contain similar tandemly repeated motifs. This encoded protein is associated with the cytoskeleton but is not involved in contraction.
Gene ID 1266
Gene name CNN3
Swiss Q15417
Synonym CNN3 Rabbit pAb,CNN3,Calponin,acidic isoform
Expression Expressed in both non-smooth muscle tissues as well as smooth muscle tissues.
Functions Thin filament-associated protein that is implicated in the regulation and modulation of smooth muscle contraction. It is capable of binding to actin, calmodulin, troponin C and tropomyosin. The interaction of calponin with actin inhibits the actomyosin Mg-ATPase activity.
