Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control HeLa,Mouse heart,Rat liver
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 26kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control HeLa,Mouse heart,Rat liver
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Mitochondrion matrix
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IP Conditions for IP
Recommended dilution 1:20 - 1:50
Suggested sample / positive control HeLa,Mouse heart,Rat liver
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 26kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background The protein encoded by this gene belongs to the peptidase family S14 and hydrolyzes proteins into small peptides in the presence of ATP and magnesium. The protein is transported into mitochondrial matrix and is associated with the inner mitochondrial membrane.
Gene ID 8192
Gene name CLPP
Swiss Q16740
Synonym CLPP,CLPP Rabbit mAb,Caseinolytic mitochondrial matrix peptidase proteolytic subunit,Endopeptidase Clp
Expression Detected in liver (at protein level). Predominantly expressed in skeletal muscle. Intermediate levels in heart, liver and pancreas. Low in brain, placenta, lung and kidney.
Functions Protease component of the Clp complex that cleaves peptides and various proteins in an ATP-dependent process. Has low peptidase activity in the absence of CLPX. The Clp complex can degrade CSN1S1, CSN2 and CSN3, as well as synthetic peptides (in vitro) and may be responsible for a fairly general and central housekeeping function rather than for the degradation of specific substrates.
