Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control LO2,HT-29,A-549
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 70kDa/75kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF Conditions for IF
Recommended dilution 1:50 - 1:200
Suggested sample / positive control LO2,HT-29,A-549
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Membrane,Multi-pass membrane protein
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ELISA Conditions for ELISA
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control LO2,HT-29,A-549
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 70kDa/75kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene is a member of the CLC family of voltage-gated chloride channels. The encoded protein is predicted to have 12 transmembrane domains, and requires a beta subunit called barttin to form a functional channel. It is thought to function in salt reabsorption in the kidney and potassium recycling in the inner ear. The gene is highly similar to CLCNKB, which is located 10 kb downstream from this gene. Multiple transcript variants encoding different isoforms have been found for this gene.
Gene ID 1187
Gene name CLCNKA
Swiss P51800
Synonym CLCNKA,CLCK1,ClC-K1,hClC-Ka,CLCNKA Antibody
Expression Expressed predominantly in the kidney. All nephron segments expressing BSND also express CLCNK proteins.
Functions Voltage-gated chloride channel. Chloride channels have several functions including the regulation of cell volume; membrane potential stabilization, signal transduction and transepithelial transport. May be important in urinary concentrating mechanisms.
