Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control U-87MG,293T,HepG2,Mouse brain,Mouse heart,Mouse kidney,Mouse liver
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 60kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF/ICC Conditions for IF/ICC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control U-87MG,293T,HepG2,Mouse brain,Mouse heart,Mouse kidney,Mouse liver
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control

Target data

Background The protein encoded by this gene is a molecular chaperone that is a member of the chaperonin containing TCP1 complex (CCT), also known as the TCP1 ring complex (TRiC). This complex consists of two identical stacked rings, each containing eight different proteins. Unfolded polypeptides enter the central cavity of the complex and are folded in an ATP-dependent manner. The complex folds various proteins, including actin and tubulin. Alternate transcriptional splice variants have been characterized for this gene. In addition, a pseudogene of this gene has been found on chromosome 8.
Gene ID 7203
Gene name CCT3
Swiss P49368
Synonym CCT3,CCT-gamma,CCTG,PIG48,TCP-1-gamma,TRIC5,CCT3 Rabbit pAb,Chaperonin containing T-complex polypeptide 1 subunit 3,hTRiC5
Functions Molecular chaperone; assists the folding of proteins upon ATP hydrolysis. As part of the BBS/CCT complex may play a role in the assembly of BBSome, a complex involved in ciliogenesis regulating transports vesicles to the cilia. Known to play a role, in vitro, in the folding of actin and tubulin.
