Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control HCT116,Rat brain
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 60kDa/72kDa/76kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF Conditions for IF
Recommended dilution 1:50 - 1:200
Suggested sample / positive control HCT116,Rat brain
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Cytoplasm,Lysosome, perinuclear region
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ELISA Conditions for ELISA
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control HCT116,Rat brain
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 60kDa/72kDa/76kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background This gene encodes a member of the glycosyl hydrolase 35 family of proteins. Alternative splicing results in multiple transcript variants, at least one of which encodes a preproprotein that is proteolytically processed to generate the mature lysosomal enzyme. This enzyme catalyzes the hydrolysis of a terminal beta-linked galactose residue from ganglioside substrates and other glycoconjugates. Mutations in this gene may result in GM1-gangliosidosis and Morquio B syndrome.
Gene ID 2720
Gene name GLB1
Swiss P16278
Synonym GLB1,EBP,ELNR1,MPS4B,Beta Galactosidase Antibody,Acid beta-galactosidase,Elastin receptor 1,Beta Galactosidase
Expression Detected in placenta (at protein level) (PubMed:8383699). Detected in fibroblasts and testis (PubMed:2511208).
Functions Isoform 1: Cleaves beta-linked terminal galactosyl residues from gangliosides, glycoproteins, and glycosaminoglycans.
