Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control U-87MG,U-251MG
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 22KDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:100
Suggested sample / positive control U-87MG,U-251MG
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Cytoplasm
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background The final step in heme metabolism in mammals is catalyzed by the cytosolic biliverdin reductase enzymes A and B (EC 1.3.1.24).[supplied by OMIM, Jul 2009]
Gene ID 645
Gene name BLVRB
Swiss P30043
Synonym BLVRB,BVRB,FLR,HEL-S-10,SDR43U1,BLVRB Rabbit pAb,Biliverdin reductase B,Biliverdin-IX beta-reductase,Green heme-binding protein,NADPH-dependent diaphorase,NADPH-flavin reductase,S-nitroso-CoA-assisted nitrosyltransferase,SCAN
Expression Predominantly expressed in liver and erythrocytes. At lower levels in heart, lung, adrenal gland and cerebrum.
Functions Broad specificity oxidoreductase that catalyzes the NADPH-dependent reduction of a variety of flavins, such as riboflavin, FAD or FMN, biliverdins, methemoglobin and PQQ (pyrroloquinoline quinone). Contributes to heme catabolism and metabolizes linear tetrapyrroles. Can also reduce the complexed Fe3+ iron to Fe2+ in the presence of FMN and NADPH. In the liver, converts biliverdin to bilirubin.
