Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500-1:2000
Suggested sample / positive control HeLa,Mouse brain
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 55kD
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:100-1:800
Suggested sample / positive control HeLa,Mouse brain
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: Cell junction,Cell projection,Cytoplasm,Cytoplasmic vesicle,Endosome,acrosome,cytoskeleton,dendrite,dendritic spine,postsynaptic cell membrane,postsynaptic density,secretory vesicle,synapse
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IF Conditions for IF
Recommended dilution 1:100-1:800
Suggested sample / positive control HeLa,Mouse brain
Key conditions Optimize fixation, permeabilization, and blocking, and include a secondary-only control
Expected result Expected fluorescence localization: Cell junction,Cell projection,Cytoplasm,Cytoplasmic vesicle,Endosome,acrosome,cytoskeleton,dendrite,dendritic spine,postsynaptic cell membrane,postsynaptic density,secretory vesicle,synapse
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IP Conditions for IP
Recommended dilution 1:50-1:200
Suggested sample / positive control HeLa,Mouse brain
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 55kD
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ELISA Conditions for ELISA
Recommended dilution 1:5000-1:20000
Suggested sample / positive control HeLa,Mouse brain

Target data

Background Master regulator of synaptic plasticity that self-assembles into virion-like capsids that encapsulate RNAs and mediate intercellular RNA transfer in the nervous system. ARC protein is released from neurons in extracellular vesicles that mediate the transfer of ARC mRNA into new target cells, where ARC mRNA can undergo activity-dependent translation. ARC capsids are endocytosed and are able to transfer ARC mRNA into the cytoplasm of neurons. Acts as a key regulator of synaptic plasticity: required for protein synthesis-dependent forms of long-term potentiation (LTP and depression (LTD and for the formation of long-term memory.
Gene ID 23237
Gene name ARC KIAA0278
Swiss Q7LC44
Synonym ARC KIAA0278;Activity-regulated cytoskeleton-associated protein (ARC/ARG3.1) (Activity-regulated gene 3.1 protein homolog) (Arg3.1);
