Applications and conditions

Where to start

These starting conditions come from the recommended dilution, validation images, and general assay practice. Titrate for your sample and detection system.

WB Conditions for WB
Recommended dilution 1:500 - 1:2000
Suggested sample / positive control Rat lung
Key conditions Use a fresh lysate, incubate at the recommended dilution, and check the predicted and observed molecular weight
Expected result Expected signal or band near 56kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control
IHC Conditions for IHC
Recommended dilution 1:50 - 1:200
Suggested sample / positive control Rat lung
Key conditions For paraffin sections, optimize retrieval buffer pH, retrieval time, and primary incubation
Expected result Expected localization: cytosol,nucleoplasm,nucleus
Controls Include a positive sample, a negative sample, and a secondary or isotype control
ELISA Conditions for ELISA
Recommended dilution See the validation images or contact technical support
Suggested sample / positive control Rat lung
Key conditions Start at the recommended dilution and titrate for your sample and assay
Expected result Expected signal or band near 56kDa
Controls Include a positive sample, a negative sample, and a secondary or isotype control

Target data

Background C2ORF13 is a component of the cellular response to chromosomal DNA single- and double-strand breaks (Iles et al., 2007 [PubMed 17353262]).[supplied by OMIM, Mar 2008]
Gene ID 200558
Gene name APLF
Swiss Q8IW19
Synonym APFL,C2orf13,PALF,Xip1,ZCCHH1,APLF,APLF Antibody,Apurinic-apyrimidinic endonuclease APLF,PNK and APTX-like FHA domain-containing protein,XRCC1-interacting protein 1
Functions Nuclease involved in single-strand and double-strand DNA break repair. Recruited to sites of DNA damage through interaction with poly(ADP-ribose), a polymeric post-translational modification synthesized transiently at sites of chromosomal damage to accelerate DNA strand break repair reactions. Displays apurinic-apyrimidinic (AP) endonuclease and 3'-5' exonuclease activities in vitro. Also able to introduce nicks at hydroxyuracil and other types of pyrimidine base damage.
